Nah, to get Megaman, you need to play 100 online matches.

That link has pics of the locations of the target markers. Even maps to give you an idea of how far along they are. But these are listed in order so you don't think "oh, I got T in Thunder, I guess that's the first one and I haven't skipped any" 'cuz you'd be wrong. Seems the map for missions 5-6 isn't working so you'll have to do without and just be able to piece it together by looking at the pics. It's no problem doing this on Easy because just about every mission you should have a TON of T-Energy. I completed just about every level with almost 9k energy (cuz I don't use energy weapons in campaign very much unless it's the Plasma Gun). So play it on easy, get all the target markers along the way, and you should have Joe and Luka unlocked in one play through. Can't believe we have to get to level 90 to get Frank West, though. I would've liked him to be wearing his default clothing from Dead Rising than have him running around in his boxers. But, eh, I'll take what I can get. Also, Joe has a helmet over his head, so you can't even see his face. Kinda disappointing. It was cool seeing Luka fight in Trial Point last night. Luka with a rocket launcher...funny stuff.
